Time to revive my favourite thread. I will be assisting Kyuu in the Tox project so you can also send me any of your queries if Kyuu is unavailable. ACONITINE Introduction: Aconitine is an Alkaloid present in the Aconitum species of Plants. Its roots are highly poisonous. Ironically, roots are often used in traditional Chinese medicine for treatment of Rheumatoid Arthritis. Structure : Chemical Formula: C34H47NO11 Appearance: Crystals LOAEL: 0.006 mg/kg LD50(GI [assumed]): 0.028 mg/kg LD50Perspective: 2-3 mg is lethal. LD50(IV mouse): 0.1 mg/kg LD50(IP mouse): 0.1 mg/kg LD50(SC mouse): 0.27 mg/kg LD50(Oral mouse): 0.1 mg/kg Perspective on LD50 of Aconite Toxicity: Aconitine is a fast acting toxin. Although it is seen that pure Aconitine crystals aren’t as toxic as raw aconite products. T1/2: 6-12 hours ADME Absorption: Gastrointestinal tract, Intravenous and Intramuscular. Distribution: Through the blood. Metabolism: Metabolised by CYP P450 enzyme in the Liver. Excretion: Urine. Mechanism of toxicity (aka: how it kills you): Information on the mechanism from Detective Conan is as follows : Aconitine is a paralytic nerve poison extracted from the leaves and roots of Aconitum species. Verdict: Correct. Technical explanation: The toxin acts on binds on voltage dependant Sodium channels present on the surface of Cardiac muscle, Skeletal Muscles and Neurons. They prolong the opening of Sodium channels, thus causing depolarisation phase to last longer causing an upstroke in the Action potential. As a result, the membrane cannot be repolarized. Translation for lay people: It allows Sodium ions to move inside the cell causing increased Sodium concentration leading to physical manifestations. Notes on accuracy in DC: Although it is true that Aconitine has neurotoxic properties, it only causes generalised muscular weakness. Its most adverse toxic effect is seen in the Heart. Death is often due to Ventricular Arrhythmia. Final note: I won’t be delving much into the treatment part but if by any 0.1% chance someone accidentally encounters such a situation, drinking strong coffee can actually hamper the progression of the poisoning. That DOES NOT mean you don’t need to go to a hospital. Something interesting: In numerous lab experiments, it has been observed that Tetrodotoxin is able to act as an antidote in Aconite poisoning since they both show mutual antagonistic effect towards each other. Of course, we don’t administer Tetrodotoxin as it is super lethal and there are way better drugs out there. But still, a poison used to kill another poison and acting like an antidote is pretty cool though :3

But not too deep: She's cried in File 1 (Case Aftermath), File 9 (Shinichi), File 16 (Case Aftermath), File 48 (Shinichi), File 71 (Shinichi), File 95 (Shinichi), File 143 (Case Aftermath), File 260 (Shinichi), File 334 (Shinichi), File 353/File 354 (Flashback – Case Aftermath), File 379/File 380 (Shinichi), File 428 (Shinichi), File 482 (Shinichi/Case Aftermath), File 650/File 651 (Shinichi), File 727 (Shinichi), File 745/File 752 (Shinichi), File 884 (Flashback – Shinichi) File 921/File 922/File 923/File 924 (Flashback – Frustration/Case Aftermath/Shinichi) and File 931/933 (Zombie Movie and Zombie encounter). Thats a total of 20 times—in a total 27 Files—12 times exclusively due to Shinichi, 4 times exclusively due to the aftermath of a case, 1 time due to Shinichi and the aftermath of a case, and 1 time when the aftermath of a case, Shinichi, and simple frustration all happened across 4 files... and then there were two instances due to zombie movie and a "zombie" encounter. I don't think I missed any, but I could be wrong about that. She cried in 6 of the 130 Files before her second suspicion arc began, cried only once during her second suspicion arc, cried 5 times between her second and third suspicion arcs, cried during her third, just one case-long, suspicion arc, and has cried 7 times since the last suspicion arc. In all, she's cried in 19 of the 273 cases, and 27 of the 945 Files.

I think Ai/Shiho harbors some affection for Conan/Shinichi, but I think she is also clever enough to see that it is one-sided. Shinichi is pretty darn hung up on Ran, to the point where he is completely blind to anyone else's attention, including Shiho's. From my understanding of Shiho's character, I don't think she will ever make her feelings known. But I think that is for the best. Recall Shiho's tragic backstory. Her parents, both members of the Black Organization, are dead. From a young age, she was shipped off to a foreign land and groomed for a special position killing people. Her beloved sister, who was probably the only person she could confide in her entire life, was killed by the Black Organization. Shiho's life was steeped in tragedy. Then she attempted suicide by taking the APTX-4869 she had developed. But instead of dying, she shrank. If any of you have known someone who seriously attempted suicide, you know they do not overcome their depression and get better right away. The depression that was strong enough to make the individual want to die still exists. It takes lots of time and lots of help for any sort of positive change. And there's also the possibility that the person will try it again, which we did see when Shiho chose to remain on the bus with the bombs. When someone is going through all of this, the pursuit of romance is probably the worst thing for them. Romance comes with such a wide array of emotions that it really isn't good for someone in such a sensitive position. Even though Shiho may have a crush on Shinichi, I think it is wise that she is not pursuing this relationship (which, given the closeness of Shinichi and Ran, would probably end in heartbreak anyway). The best way for Shiho to recover is to take time to relax away from her regular stresses, spend time with her new found friends, and discover what makes her happy. I think Shiho's shrinking incident was actually a blessing. She got away from the Black Organization, she made some friends, and she is finally starting to live a normal life. Because Haibara was never really able to have a childhood, I can see her choosing to stay a child at the end while Shinichi chooses to grow up. I think this would be an ideal way for Shiho to reboot her life, and I think it's clear that this current lifestyle is already making her much happier. So there you have it. I might have gone off-topic a bit, but my main reason I disagree with Conan/Ai is because I don't see that scenario as being healthy for Shiho. Although everyone has their own opinion, I hope mine makes sense to you guys! <3
